چکیده
Topiramate is an anti-epileptic drug that commonly prescribed not just to prevent seizures but also migraine headaches, with over 8 million prescriptions dispensed annually. use during pregnancy has been linked significantly increased risk of babies born orofacial clefts (OFCs). However, the exact molecular mechanism topiramate teratogenicity unknown. In this study, we first used unbiased antibody array analysis test effect on human embryonic palatal mesenchyme (HEPM) cells. This identified 40 differentially expressed proteins, showing strong connectivity known genes associated clefts. among OFC genes, only TGFβ1 was upregulated in analysis. Next, validated could increase expression and downstream target phospho-SMAD2 primary mouse (MEPM) Furthermore, showed treatment MEPM cells SOX9. SOX9 overexpression chondrocytes cause cleft palate mouse. We propose mediates upregulation signaling through activation γ-aminobutyric acid (GABA) receptors palate. play critical roles morphogenesis, their abnormal provides a plausible etiologic for teratogenic effects topiramate.

Generation of an immortalized mouse embryonic palatal mesenchyme cell line
Palatogenesis is a complex morphogenetic process, disruptions in which result in highly prevalent birth defects in humans. In recent decades, the use of model systems such as genetically-modified mice, mouse palatal organ cultures and primary mouse embryonic palatal mesenchyme (MEPM) cultures has provided significant insight into the molecular and cellular defects underlying cleft palate. Howev...
متن کاملMetanephric mesenchyme contains embryonic renal stem cells.
Renal epithelial cells derive from either cells of the metanephric mesenchyme or ureteric bud cells, but the origin of other renal cells is unclear. To test whether metanephric mesenchymal cells generate cells other than epithelial, we examined the developmental potential of a metanephric mesenchymal cell line (7.1.1 cells) and of primary cultures of metanephric mesenchymal cells. 7.1.1 Cells e...
متن کاملCharacterization and evaluation of the efficacy of cationic complex mediated plasmid DNA delivery in human embryonic palatal mesenchyme cells.
The purpose of this study was to develop and test a non-viral gene delivery system that can be employed to deliver genes of interest into a pre-osteoblastic cell line. Human embryonic palatal mesenchymal (HEPM 1486) cells were transfected with vector-plasmid DNA (pDNA) complexes. We explored calcium phosphate and polyethylenimine (PEI) as non-viral vectors and compared their respective in vitro...
متن کاملComparing the Expression Levels of Alkaline Phosphatase, Gfra1, Lin28, and Sall4 Genes in Embryonic Stem Cells, Spermatogonial Stem Cells, and Embryonic Stem-Like Cells in Mice
Background and purpose: Spermatogenesis is a well-organized process that is influenced by a variety of factors. Alkaline phosphatase, and Gfra1, Lin28, and Sall4 genes are among the key players in this interconnected process. This study aimed to investigate the expression levels of Gfra1, Lin28, and Sall4 genes in embryonic, spermatogonial, and embryonic stem-like (ES-like) cells in mice. Mate...
متن کاملImplications of TGFβ on Transcriptome and Cellular Biofunctions of Palatal Mesenchyme
Development of the palate comprises sequential stages of growth, elevation, and fusion of the palatal shelves. The mesenchymal component of palates plays a major role in early phases of palatogenesis, such as growth and elevation. Failure in these steps may result in cleft palate, the second most common birth defect in the world. These early stages of palatogenesis require precise and chronolog...
